چکیده

www.pharmtech.com n the manufacture of many pharmaceutical products, dry-particle blending is a critical step that directly affects content uniformity. Tumbling blenders—a common way to mix granular constituents—are hollow containers attached to a rotating shaft; the vessel is partially loaded with materials and rotated. Their main advantages are large capacities, low-shear stresses, flexibility, containment, and easy cleaning. The blenders come in a variety of geometries and sizes (ranging from a minimum of ,16 qt to a maximum .500 ft). The convective blender is another common mixer, in which flow is created by impellers rotating within a fixed shell. The blender can impart high shear, reduce ingredient segregation, and be used for wet granulation. Although the blenders are available in several sizes, the largest capacity is often an inverse function of the maximum shear rate. Currently, no rigorous techniques can predict blending scale-up criteria for either type of blender without prior experimental work. Typically, blending studies start with a smallscale, try-it-and-see approach, during which the following questions usually arise: Scale Up of PowderBlending Operations

Characterization of pharmaceutical powder blends by NIR chemical imaging.

This study demonstrates the capabilities of NIR imaging as an effective tool for characterization of pharmaceutical powder blends. The powder system used in this small-scale powder blending study consists of acetaminophen (APAP, the model API), microcrystalline cellulose (MCC) and lactose monohydrate. Mixtures of these components were blended for different times for a total of ten time points (...

Effects of rotation rate, mixing angle, and cohesion in two continuous powder mixers—A statistical approach

a r t i c l e i n f o Keywords: Continuous powder mixing Statistical analysis ANOVA Homogeneity Residence time In this paper we examine the effect of rotation rate, mixing angle, and cohesion on the powder residence time and the content uniformity of the blend exiting from two continuous powder mixers. In addition, differences in mixing performance between the two blenders are examined. Analysi...

Scale-up strategy for continuous powder blending process

a r t i c l e i n f o Continuous powder mixing has attracted a lot of interest within the pharmaceutical industry. Much work has been done recently that targets the characterization of continuous powder mixing. In this paper, a quantitative scaling up strategy is introduced that allows the transition from lab to industrial scale. The proposed methodology is based on the variance spectrum analys...

The Scale Method for Blending Operations in Functionally-Based Constructive Geometry

This paper presents a scale method for developing high dimensional scale functions to blend implicitly defined objects. Scale functions are differentiable on the entire domain except the origin, provide blending range control, and behave like Min/Max operators everywhere, so even a successive composition of blending operations containing overlapped blending regions can be generated smoothly. Be...

Flowsheet Modeling of a Continuous Direct Compression Tableting Process at Production Scale

A flowsheet model for a continuous direct compression tableting process is developed and applied to process data. The model contains powder feeding, blending and tablet compaction unit operations. The dataset was provided by Merck & Co., Inc., Kenilworth, NJ USA using a GEA ConsiGma continuous direct compression process designed for a 50 kg/h throughput using a six ingredient, potentially comme...
